I have mid-size to large hands and plan to use this on a desktop so, it need not be "portable". I don't like tiny mice. I've heard good things about the Logitech MX1000, but $80 bucks for a mouse?!
Is laser that much better than the regular optical mouse?

Since you're asking for the "best", I have to mention the Razer Mamba, but if you think $80 is steep, you're probably not going to like the Mamba's $130 price tag.
Short of that, I recommend the Logitech G7. -
If i ever move to wireless somday, this would be my choice Logitech Performance Mouse MX, if you find it too expensive maybe Logitech Anywhere Mouse MX.

Mx1000 is quite old now. A fantastic mouse though, nice, big and chunky that you can use either fingers or hand to hold.
Still, I am sure logitech has something newer out although not sure on better. -

Do you plan on gaming, or is it more for generalized usage?
I have several mice..they are very comfy..but mainly Notebook mice.

Just generalized usage, I don't game. I'm really worried about getting a mouse that's too small since I've heard that it gives cramps. Anyone know any good deals?!

General usage definitely needs the Performance MX series. I have had/used both the MX Revolution (desktop) and VX Nano (laptop) series mice and they are some of the best mice that I have ever used (in a NON gaming capacity of course). The comfort level on the MX series is just amazing. I am at my PC more than 14 hours a day usually (work from home) so I need a good comfortable mice to work with. My MX Rev just died so I am probably going to replace it with a P MX. Either way though you won't find a better wireless mouse.

The MX Revo (and the Performance MX by allusion) has the joint-best shape I've had in a full-sized mouse. The MX1000 is also a very unfatiguing shape for me and these are my best mice to date.
I did find the VX Revolution a superb mousing shape as well for a more compact mouse, but it doesn't seem up to the same duty cycle as the MX. -
Why a wireless mouse for desktop use? I use a wireless trackball at home and it is so annoying having to swap batteries frequently. There are rechargeable mice for desk use that would help in this regards, but after a few years the battery would probably die and then the mouse would be useless. A good wired mouse would last a lot longer than this.
