    My Fn keys on my Asus have been working fine until one day they suddenly stopped working.
    Could anyone tell me why or how to enable them please?

    Check in BIOS, there may be an option there to enable/disable.

    If not, do a system restore back to a day it worked.

    Maybe easier, remove all the ATK drivers and utilities, download the latest from support.asus.com, and install them.
