I'm looking to upgrade to Vista Ultimate from Home Premium. Is there a guide on that?

I don't think you need a guide for it. If you haven't purchased the software, you can take a look at Windows Anytime Upgrade from Home to Ultimate. It should cost around 189USD.
The drivers used in Home Premiums are directly compatible with Ultimate, unless you're switching from a 32bit operating system to a 64bit one. -

I'm an uber noob, so this may be a stupid question. When upgrading, does it wipe everything off that has been previously installed? Do I need to re-install all programs / drivers / etc.? -
It depends. If I remember correctly, if you're only upgrading your Vista to a higher version, you do NOT need to backup programs and drivers. However, just for the sake of safety, you might want to back them up.
