I don't have the answer for the sound problem, but I can point you in the right direction. Google is your friend.

Anyway, there's a setting somewhere that allows you to force your computer to play both through headphones and the speakers. Yours looks like it's enabled.

Check your Realtek audio manager. There should be a setting that allows you to disable your laptop speakers when the audio out jack is in use.

Try reinstalling your audio driver. Check the Asus website to see if there's a newer version.

I have a sound problem myself. I have the G50v Best Buy Ver. When I do a speaker test in Vista sound control panel I get sound out of the right laptop speaker for both left and right channel test(left channel on right speaker, right channel on right speaker). No sound out of left speaker, ever, with any apps. The balance is ok and the speaker setup is set to stereo. When I plug headphone into the headphone the sound is ok (left channel on left speaker, Right channel on right speaker). Any ideas?

Btw. When laptop boots the "republic of gamers" sound effect is also on right side speaker only im pretty sure. So it may not be a vista issue.

I solved mine, i googled it and found that I'm not alone :)
it was the audio jack prob, it has something in it that mutes the int. speakers when headphones inserted into the jack, so I played with the jack(lol) then the prob solved! I think that thing was stuck at it place.

That's strange. Not working at POST suggests hardware problem. Working in control panel suggests software problem.

For a hardware problem, I'd suggest opening up the notebook to see if the speaker wires are properly connected. We have a disassembly manual on the Info Booth. Make sure you don't remove heatsinks.

For a software problem I would suggest a driver reinstall.

Because of the possibility of both, I don't know what to suggest. :D
