If I purchase a graphic card that has s-video, vga and dvi port - can I use more than one port at the same time or will it only output to one at a time?

Yes and yes.
You can configure however you like. But your options are an extended display or mirrored display. -

If I connect two external displays my laptops screen turns off which is normal since most gfx cards support two displays.

My laptop cannot display via the vga out and s-video out at the same time, but I was hoping that with a desktop graphic card, it can display the s-video to one tv and the dvi to another tv, with no lcd monitor. In clone (or mirrored) mode.

You are likely able to have one analogue and one digital connection at any time.
