I have a regular Nintendo DS, when I first got it a long time ago I read something that if you have a wireless router you can hook up to it with the DS and play against people with the DS over your internet connection. Is this true?

Yeap, the DS wifi is just like a notebook wifi, you connect to it to play online.
I do it all the time on my DSLite.
The USB Nintendo wireless adapter is for people who don't have a router, and that adapter acts as a 'router' for the DS. -
I've done it with a secured Linksys router, but I can't get it to connect to my belkin because its security is to high. I played Mario Cart DS online with other people, its pretty fun. I would get the USB wireless adapter but it costs around $50(At least thats how much it was a while ago, haven't checked since)

To hook it up to your wirless network? No you just go to the DS WiFi settings in the game and tell it to search for an access point. if you have securiteis on it then you can put the MAC of your DS into your router and and all that. I think you get your MAC from the WiFi settings on your DS.
If you have one of the Nintendo USB access points then just install the CD that comes with it and it will work.
