    Does anyone know if it's possible to replace the Intel 4965AGN with a Intel 5300 in a Lenovo T61?

    I think you can do it as long as you get a Lenovo-branded 5300. Not 100% sure of that, though; afaik, Lenovo's got this whitelisting thing going on which isn't very upgrade friendly.

    Will it make much of a difference? Not in my opinion unless you've got a specific reason to do so.

    4965AGN is a good card --there's very little reason to upgrade. Unless you have some reason to use Intel's MyWiFi technology, you're going to spend money you're unlikely to recoup in performance, or range.

    The reason to upgrade is if you're having an issue with the card. If not, it's worth the hassle, my opinion of course.
