Yes, you will need to install Windows on your new hard drive. Also, there really isn't a solid way to transfer applications from hard drive to hard drive yet, so you'll need to install those too. You files are easily transferable.

You can’t simply copy programs and Windows on to a new hard disk. However, you can make a disk image of the old Windows partition (including the MBR) using a tool like Norton Ghost or Acronis, then extract that on to the new HDD. This way you can transfer Windows and programs without reinstalling, but it can be a lot messier. Therefore I also recommend a new installation.

Of course settings and files can be transferred manually via a number of was but geez, HD Cloning is so simple and fast its definitely worth the price of quality software.
PS I used Acronis to transfer off my stock Fujitsu 120gig 5400 rpm drive to a Hitachi 200gig 7200 rpm drive. Easy pesy.
