Planning to upgrade to a SSD soon. Question is, does the mSATA port in the X220t have any speed limitations on the interface when compared to a SSD in the 2.5" bay?

yes, it is SATA 2 (like all mSATA ports afaik). The one in the bay is Sata 3.

If you don't need the extra space provided by an mSATA + HDD combo then there is little point in going mSATA over 2,5" SSD (faster, cheaper, easier to install).
I have 256GB mSATA + 1TB HDD - amazing for a 12,5" Laptop (and sadly not possible in the X240)

Unless you're doing stuff that puts a heavy load on the controller, typical notebook usage does not do this, there's little practical difference between SATA II and III because it's the latency, not the throughput, that makes SSDs fast.
