No - it should be fairly easy to access and replace your notebook's HD and you shouldn't have to disassemble the entire chasis to replace it either. Just make sure that your new HD drive is compatable with your older one and you should be fine.

Since we are on the topic of upgradeability, does anyone know if the P105-s9312’s core duo can be upgraded to core 2 duo? Also, regarding the hard drive upgrade, how would one go about finding a compatible hard drive? Or, more specifically, a 80-100 gig 7,200 rpm hard drive upgrade.

The hard drive is a 2.5" SATA for notebooks.
Here is a list of all 7200RPM drives that will work from Newegg.com

It isn't certain if you can upgrade or not since the upgrade is highly dependent upon the release of the maker releasing a new BIOS within the same model (Toshiba would have to release a P100 notebook with a Core Duo without messing with the insides). For the HD: It's up to you to figure out whether your notebook uses ATA-6, PATA, or SATA. I'm pretty sure the P100 would have a SATA drive.
