Try re-installing Sony Shared Library and Sony Notebook Utilities. If your model doesn't have the latter, look for Setting Utility Series and VAIO Event Service (these are included in the Notebook Utilities). You should be able to find these in Vaio Care (Recovery -> Reinstall apps) or your Sony support site.

The most important one of the list jpride gave is VAIO Event Services. It's what makes the Fn, ASSIST, and S1 keys point to their respective funtions.

Shared Library and Notebook Utilities, though Sony insists they're required, are not installed on my system and my other VAIO functions work fine. I'd let them be installed if I knew what they did, but what I can find online equates to folklore, since Sony doesn't detail their functions.
