Does it make a difference for the most accurate calibration result if you start out with a fully loaded battery or partial loaded one ?

If by calibration you mean that you want to force the battery's smart chip to recalculate the actual Wh it's able to hold, then all you should need to do is run the battery down to 0% and then charge it back, uninterrupted, to 100%. It doesn't matter if you start fully charged or not.

It's a bad idea to do this more often than every 3-6 months. Frequently dropping your battery below 30% capacity will decrease the battery's capacity to hold a charge.
