I've got about 13 GB in my Downloads library on 64-bit Win 7, on a 160 GB Intel X-25M SSD with almost 50 GB free space, and it takes about a minute for the library to be accessed the first time I open it every boot.
I haven't noticed this problem with any other libraries or folders. About 85% of the contents of the Downloads library are audio files in a folder with about 40 subfolders. Any ideas what might be going wrong here?

Do you have any external drives connected to the machine? We have seen this kind of behavior in many machines with an external source that Windows is having a hard time reading to add to the list it is trying to present to you.

right click the folder - properties- optimize this library for - Mine is set to documents and opens very fast (6 GIG assorted content on SSD)
You could try general items, or music ... just to test also -

I have a couple external drives that I hook up from time to time, but the last few times it's happened there's not been any external storage set up.
The library was optimized for general items. I've set it to music (since it's primarily audio files for now). We'll have to see upon my next restart whether or not that helps, if not I'll give documents a try.
Incidentally, the big chunk of audio files predates the loading problem by a couple months. Not sure if that means anything, though.
Downloads library slow to open on SSD
