I have the MSI Titan GT80S SLI with the following configuration:

    512GB SSD M.2 2280: Windows 10, primary boot disk
512GB SSD M.2 2280 : Wndows 10 clone disk of the above, used as a backup boot disk
1TB SSD M.2 2280: - Linux to be installed here
    1TB HDD: Windows data disk

For some reason, the 3rd M.2 2280 SSD does not appear as a bootable device at all. In BIOS. How does Grub get configured to point to this third SSD once Linux installed on it?

Does it come with @Prema BIOS? If so, you can disable other drives and enable only the disk you want to install Linux.
I'd windows partitioning to create 3 partitions:
    1. one 500MB for grub efi aka ESP
    2. Linux root partition
    3. page file/swap set to 512MB
You need to install bootloader to ESP to work correctly.
