    1) Only the Ultra-fast booting option should be used right?

    2) What defragmenter does tunexp use after rearranging the files? The vista defragmenter or does it have its own defragmenter?

    TuneXP was designed for XP!

If you want a good program for Vista 64 get a program like Vista Manager.

Do NOT use TuneXP on Vista, you can seriously mess up your system. Much analysis has been done (mostly by Gary) in the Vista Tweaks thread ( link) as to what Ultra-fast booting function TuneXP does so it can be replicated in Vista. Gary found that it just uses Windows' built-in defragmenter using a switch to focus on the boot files instead of the whole drive.

Gary wrote a batch file that does this in Vista, and can be found in the thread I linked above (step 16).
