What is the fastest and best processor I can upgrade my e380 aspire desktop to, I am looking to get the most of this computer and the processor is something I have yet to upgrade. Can I use a tri/quad core? would a AMD Phenom 8650 work? or would i have to end up buying a new motherboard?

Tough to say, the chipset should be the 6150 which should support the Phenom but it's up to Acer to release a BIOS which does. Not sure if the current ones have that support built-in.
